The R160H variant has not been published in association with CPVT, however, it has been reported as a variant of uncertain significance by another clinical laboratory in ClinVar (SCV000197206.3; Landrum et al., 2016). The R160H variant was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. Additionally, in silico analysis suggests this variant is probably damaging to the protein structure/function. While this substitution occurs at a position that is conserved across most species, Histidine is the wild type in the lizard. Finally, the R160H variant is a conservative amino acid substitution, which is not likely to impact secondary protein structure as these residues share similar properties.Therefore, based on the currently available information, it is unclear whether this variant is pathogenic or rare benign. - |
