Charcot-Marie-Tooth disease type 2 Pathogenic:1
This sequence change replaces threonine, which is neutral and polar, with arginine, which is basic and polar, at codon 528 of the LMNA protein (p.Thr528Arg). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individuals with autosomal dominant Emery-Dreifuss muscular dystrophy (PMID: 14684700, 20376791, 26165385). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 66850). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt LMNA protein function with a positive predictive value of 95%. For these reasons, this variant has been classified as Pathogenic. -
