Mutations in this gene cause variegate porphyria, an autosomal dominant disorder of heme metabolism resulting from a deficiency in protoporphyrinogen oxidase, an enzyme located on the inner mitochondrial membrane. The observation of one or more missense substitutions at this codon (p.Gly11Asp and p.Gly11Ser) in affected individuals suggests that this may be a clinically significant residue (PMID: 21048046, 16947091, 11348478). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). This variant has not been reported in the literature in individuals with PPOX-related disease. This variant is not present in population databases (ExAC no frequency). This sequence change replaces glycine with cysteine at codon 11 of the PPOX protein (p.Gly11Cys). The glycine residue is highly conserved and there is a large physicochemical difference between glycine and cysteine. -
