This variant was detected in a female with delayed and imbalanced intellectual and motor development, growth retardation, borderline macrocephaly, hirsutism. The variant was confirmed to be of a de novo origin. Rare variants altering the canonical donor splice sites in the PBX1 gene are well documented as a molecular cause of congenital anomalies of kidney and urinary tract syndrome with or without hearing loss, abnormal ears, or developmental delay (CAKUTHED) (OMIM:617641) (PMID:37571997;28270404;29036646;28566479). To conclude, the variant is classified as pathogenic (ACMG PVS1, PM2, PS2). -
