Genes affected
ASPM (HGNC:19048): (assembly factor for spindle microtubules) This gene is the human ortholog of the Drosophila melanogaster 'abnormal spindle' gene (asp), which is essential for normal mitotic spindle function in embryonic neuroblasts. Studies in mouse also suggest a role of this gene in mitotic spindle regulation, with a preferential role in regulating neurogenesis. Mutations in this gene are associated with microcephaly primary type 5. Multiple transcript variants encoding different isoforms have been found for this gene.[provided by RefSeq, May 2011]

Pathogenic, criteria provided, single submitter | clinical testing | GeneDx | May 09, 2014 | The c.1959_1962delCAAA mutation in the ASPM gene has been reported previously in association with primary microcephaly (Bond et al., 2003). The deletion of four bases causes a frameshift starting with codon Asparagine 653, changes this amino acid to a Lysine residue and creates a premature Stop codon at position 14 of the new reading frame, denoted p.Asn653LysfsX14 (N653KfsX14). This mutation is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. - |
