Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 5.6e-05 in 251302 control chromosomes. This frequency is not significantly higher than estimated for a pathogenic variant in USH2A causing Usher Syndrome (5.6e-05 vs 0.011), allowing no conclusion about variant significance. c.11156G>A has been reported in the literature in multiple individuals affected with USH2A-related disorders (e.g., Chen_2014, Yang_2015, Gao_2021), and the variant has been shown to segregate with disease in affected family members. These data indicate that the variant is very likely to be associated with disease. 13 ClinVar submitters (evaluation after 2014) have cited the variant with conflicting assessments. Five submitters classified the variant as pathogenic, 7 submitters classified the variant as likely pathogenic, and one submitter classified the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as pathogenic. -
