ACTA1 (HGNC:129): (actin alpha 1, skeletal muscle) The product encoded by this gene belongs to the actin family of proteins, which are highly conserved proteins that play a role in cell motility, structure and integrity. Alpha, beta and gamma actin isoforms have been identified, with alpha actins being a major constituent of the contractile apparatus, while beta and gamma actins are involved in the regulation of cell motility. This actin is an alpha actin that is found in skeletal muscle. Mutations in this gene cause a variety of myopathies, including nemaline myopathy, congenital myopathy with excess of thin myofilaments, congenital myopathy with cores, and congenital myopathy with fiber-type disproportion, diseases that lead to muscle fiber defects with manifestations such as hypotonia. Therefore, this variant has been classified as Likely Pathogenic. Variants that disrupt the p.Lys375 amino acid residue in ACTA1 have been observed in affected individuals (PMID: 11525890, 19562689, 15236405, 15138616). This suggests that it is a clinically significant residue, and that other variants that disrupt this residue are likely to be causative of disease. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). This variant has been observed to be de novo in an individual affected with nemaline myopathy (PMID: 15336687). This variant has also been reported as p.Lys373Glu. This variant is not present in population databases (ExAC no frequency). This sequence change replaces lysine with glutamic acid at codon 375 of the ACTA1 protein (p.Lys375Glu). The lysine residue is highly conserved and there is a small physicochemical difference between lysine and glutamic acid. -
