Genes affected
HNRNPR (HGNC:5047): (heterogeneous nuclear ribonucleoprotein R) This gene encodes an RNA-binding protein that is a member of the spliceosome C complex, which functions in pre-mRNA processing and transport. The encoded protein also promotes transcription at the c-fos gene. Alternative splicing results in multiple transcript variants. There are pseudogenes for this gene on chromosomes 4, 11, and 10. Uncertain significance, criteria provided, single submitterclinical testingAmbry GeneticsFeb 23, 2022The c.919_920dupCA (p.Q307Hfs*7) alteration, located in exon 8 (coding exon 7) of the HNRNPR gene, consists of a duplication of CA at position 919, causing a translational frameshift with a predicted alternate stop codon after 7 amino acids. This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. However, loss-of-function of HNRNPR has not been established as a mechanism of disease. This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
