To our knowledge, functional studies have not been performed for this variant. In a large international case-control study, this variant was reported in 2/60466 breast cancer cases and 4/53461 controls (OR=0.442, 95%CI 0.081 to 2.414, p-value=0.429; PMID: 33471991). This variant has been identified in 7/282722 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -

The p.P430L variant (also known as c.1289C>T), located in coding exon 13 of the MUTYH gene, results from a C to T substitution at nucleotide position 1289. The proline at codon 430 is replaced by leucine, an amino acid with similar properties. This amino acid position is well conserved in available higher vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. -
