This missense variant replaces proline with leucine at codon 430 of the MUTYH protein. This variant is also known as c.1247C>T (p.Pro416Val) based on an alternative transcript (NM_001048171). Computational prediction is inconclusive regarding the impact of this variant on protein structure and function (internally defined REVEL score threshold 0.5 < inconclusive < 0.7, PMID: 27666373). To our knowledge, functional studies have not been performed for this variant. In a large international case-control study, this variant was reported in 2/60466 breast cancer cases and 4/53461 controls (OR=0.442, 95%CI 0.081 to 2.414, p-value=0.429; PMID: 33471991). This variant has been identified in 7/282722 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -

The p.P430L variant (also known as c.1289C>T), located in coding exon 13 of the MUTYH gene, results from a C to T substitution at nucleotide position 1289. The proline at codon 430 is replaced by leucine, an amino acid with similar properties. This amino acid position is well conserved in available higher vertebrate species.
