Variant summary
Our verdict is Pathogenic. The variant received 18 ACMG points: 18P and 0B. PVS1PM2PP5_Very_Strong
The NM_000329.3(RPE65):c.825C>G(p.Tyr275*) variant causes a stop gained change. The variant was absent in control chromosomes in GnomAD project. In-silico tool predicts a pathogenic outcome for this variant. Variant has been reported in ClinVar as Pathogenic (★★). Synonymous variant affecting the same amino acid position (i.e. Y275Y) has been classified as Likely benign. Variant results in nonsense mediated mRNA decay.

Leber congenital amaurosis 2;C3151086:Retinitis pigmentosa 20 Pathogenic:1
This sequence change creates a premature translational stop signal (p.Tyr275*) in the RPE65 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in RPE65 are known to be pathogenic (PMID: 9326941, 9501220, 9843205, 18632300). This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individuals with inherited retinal dystrophy (PMID: 30576320, 31273949, 31630094). ClinVar contains an entry for this variant (Variation ID: 587393). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. For these reasons, this variant has been classified as Pathogenic.
