This variant has not been reported in a large population database, indicating this variant is rare. At this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence. -

The p.P140L variant (also known as c.419C>T), located in coding exon 2 of the RBM20 gene, results from a C to T substitution at nucleotide position 419. The proline at codon 140 is replaced by leucine, an amino acid with similar properties. This alteration has been reported in an ostensibly healthy control cohort (Mazzarotto F et al. Circulation, 2020 Feb;141:387-398). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
