This sequence change affects codon 977 of the RET mRNA. It is a 'silent' change, meaning that it does not change the encoded amino acid sequence of the RET protein. This variant is present in population databases (rs375414982, gnomAD 0.006%). This variant has not been reported in the literature in individuals affected with RET-related conditions. ClinVar contains an entry for this variant (Variation ID: 579590). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. This synonymous variant causes a nucleotide substitution but does not change the encoded amino acid at codon 977 in the RET protein. Splice site prediction tools suggest that this variant may impact RNA splicing by creating a cryptic splice donor site. The c.2931C>T variant (also known as p.S977S), located in coding exon 17 of the RET gene, results from a C to T substitution at nucleotide position 2931. This nucleotide substitution does not change the serine at codon 977. This nucleotide position is poorly conserved in available vertebrate species. However, in silico splice site analysis predicts that this alteration will result in the creation or strengthening of a novel splice donor site. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
