The p.M1? pathogenic mutation (also known as c.1A>T) is located in coding exon 1 of the SDHD gene and results from a G to A substitution at nucleotide position 3. This alters the methionine residue at the initiation codon (ATG). This variant was identified in an individual with pheochromocytoma/paraganglioma from the TCGA cohort (Huang KL et al. Cell, 2018 04;173:355-370.e14). Different alterations disrupting this initiation codon (including c.3G>A, c.3G>C, and c.2T>A) have been observed in multiple individuals with pheochromocytomas and/or paragangliomas (Cascón A et al. J. Clin. Endocrinol. Metab. 2009 May;94(5):1701-5; Ben Aim L et al. J. Med. Genet. 2019 Aug;56(8):513-520; Badenhop RF et al. Genes Chromosomes Cancer. 2001 Jul;31(3):255-63; Lee SC et al.
