The encoded protein is one of two integral membrane proteins anchoring the complex to the matrix side of the mitochondrial inner membrane. Mutations in this gene are associated with the formation of tumors, including hereditary paraganglioma. Transmission of disease occurs almost exclusively through the paternal allele, suggesting that this locus may be maternally imprinted. Likely pathogenic, criteria provided, single submitter | clinical testing | Ambry Genetics | Jun 12, 2024 | The c.278_280dupATT variant (also known as p.Y93dup), located in coding exon 3 of the SDHD gene, results from an in-frame duplication of ATT at nucleotide positions 278 to 280. This results in the duplication of an extra residue between codons 93 and 94. This variant was detected in individuals with carotid body tumors (Ambry internal data). Based on internal structural assessment, this alteration disrupts the fold of the SDHD protein (Sun F et al. Cell, 2005 Jul;121:1043-57). In addition, this alteration is predicted to be deleterious by in silico analysis (Choi Y et al. PLoS ONE. 2012;7:e46688). Based on the majority of available evidence to date, this variant is likely to be pathogenic. - |
