Inborn genetic diseases Uncertain:1
The c.13C>A (p.P5T) alteration is located in exon 3 (coding exon 1) of the CEP164 gene. This alteration results from a C to A substitution at nucleotide position 13, causing the proline (P) at amino acid position 5 to be replaced by a threonine (T). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
Nephronophthisis 15 Uncertain:1
ClinVar contains an entry for this variant (Variation ID: 1060292). This sequence change replaces proline, which is neutral and non-polar, with threonine, which is neutral and polar, at codon 5 of the CEP164 protein (p.Pro5Thr). This variant is present in population databases (rs749869599, gnomAD 0.02%). This variant has not been reported in the literature in individuals affected with CEP164-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
