The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.316-96G>A in individuals affected with Hemoglobinopathy and no experimental evidence demonstrating its impact on protein function have been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. There are several reported disease variants described in this intron (e.g., c.316-3, 316-7, 316-12, 316-14, 316-106, 316-146, and 316-197) that generate an alternative splice-site, suggesting this deep intronic variant should be interpreted with caution.
