Variant summary
Our verdict is Uncertain significance. Variant got 0 ACMG points: 2P and 2B. PM2BP4_Moderate
The NM_000518.5(HBB):c.92+6T>A variant causes a splice region, intron change involving the alteration of a non-conserved nucleotide. The variant was absent in control chromosomes in GnomAD project. In-silico tool predicts a benign outcome for this variant. 2/3 splice prediction tools predict no significant impact on normal splicing. Variant has been reported in ClinVar as Uncertain significance (★).

The HBB c.92+6T>A variant, to the best of our knowledge, has not been reported in the published literature. It also has not been reported in large, multi-ethnic general populations (Genome Aggregation Database, http://gnomad.broadinstitute.org). A different variant at the same position, c.92+6T>C, interferes with beta-globin mRNA splicing and is associated with beta(+)-thalassemia. Based on the available information, we are unable to determine the clinical significance of the c.92+6T>A variant. Clinical correlation and genetic counseling are recommended. -
