This changes the amino acid from a glutamic acid to a stop codon within coding exon 1. This alteration has been reported in four individuals with multiple endocrine neoplasia type 1 (MEN1), all of whom were reported to have non-functioning adrenal tumors (Waldmann J et al. Langenbecks Arch Surg, 2007 Jul;392:437-43). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. - |
