The p.M275L variant (also known as c.823A>C), located in coding exon 7 of the CDK4 gene, results from an A to C substitution at nucleotide position 823. The methionine at codon 275 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. -
