The encoded preproprotein is proteolytically processed following assembly into large multimeric complexes. Analysis of this variant using bioinformatics tools for the prediction of the effect of amino acid changes on protein structure and function yielded predictions that this variant is damaging. This variant is reported in ClinVar (Variation ID: 100616) and is found in the African/African-American population with an allele frequency of 0.08% (20/24,896 alleles) in the Genome Aggregation Database (v2.1.1). Computational analyses predict that this variant is deleterious (REVEL: 0.907). Due to conflicting information, the clinical significance of this variant is uncertain at this time.
