The proline at codon 190 is replaced by alanine, an amino acid with highly similar properties. To our knowledge this exact alteration has not been reported in the literature, however an alteration at the same codon, p.P190L (c.569C>T), has been reported in a Greek individual with breast cancer at age 55 along with a family history of a sister with breast cancer at age 55, and a paternal aunt with breast cancer (Konstantopoulou I et al, Clin. Genet. 2014 Jan; 85(1):36-42). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
