The p.P190A variant (also known as c.568C>G), located in coding exon 6 of the BRCA2 gene, results from a C to G substitution at nucleotide position 568. The proline at codon 190 is replaced by alanine, an amino acid with highly similar properties. To our knowledge this exact alteration has not been reported in the literature, however an alteration at the same codon, p.P190L (c.569C>T), has been reported in a Greek individual with breast cancer at age 55 along with a family history of a sister with breast cancer at age 55, and a paternal aunt with breast cancer (Konstantopoulou I et al, Clin. Genet. 2014 Jan; 85(1):36-42). This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
Hereditary breast ovarian cancer syndrome Uncertain:1
This sequence change replaces proline, which is neutral and non-polar, with alanine, which is neutral and non-polar, at codon 190 of the BRCA2 protein (p.Pro190Ala). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with BRCA2-related conditions. ClinVar contains an entry for this variant (Variation ID: 219928). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt BRCA2 protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
