PM2_Supporting, BP1_Strong c.2491G>T, located in exon 11 of the BRCA2 gene, is predicted to result in the substitution of Val by Phe at codon 831, p.(Val831Phe). This position is outside a (potentially) clinically important functional domain and, moreover, the SpliceAI algorithm predicts no significant impact on splicing (BP1_strong). It is not present in the population database gnomAD v2.1.1, non cancer dataset (PM2_supporting). To our knowledge, neither relevant clinical data nor well-stablished functional studies have been reported for this variant and there are no reports of pathogenic missense variants in the same codon. Also, it has not been reported neither in ClinVar, BRCA Exchange nor in LOVD databases. Based on currently available information, the variant c.2491G>T should be considered a likely benign variant according to ClinGen-BRCA2 Guidelines version v1.0.0. -
