This missense variant replaces methionine with threonine at codon 1168 of the BRCA2 protein. Computational prediction is inconclusive regarding the impact of this variant on protein structure and function (internally defined REVEL score threshold 0.5 < inconclusive < 0.7, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in a family with a history of breast and/or ovarian cancer (PMID: 16826315). This variant has been identified in 1/250868 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
The p.M1168T variant (also known as c.3503T>C), located in coding exon 10 of the BRCA2 gene, results from a T to C substitution at nucleotide position 3503. The methionine at codon 1168 is replaced by threonine, an amino acid with similar properties. This alteration has been identified in individuals diagnosed with breast and/or ovarian cancer (Peixoto A et al. Fam. Cancer. 2006 Jul;5:379-87; Trujillano D et al. J Mol Diagn. 2015 Mar;17:162-70; Boga I et al. Eur J Breast Health, 2023 Jul;19:235-252). This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
