The normal sequence, with the bases that are deleted in braces and inserted in brackets, is TGTT[GA][AT]AGGT. This in frame deletion and insertion, also denoted BRCA2 6643_6644delGAinsAT using alternate nomenclature, occurs on the same allele (in cis) and results in the missense change of a Glutamic Acid to an Isoleucine (GAA>ATA). This variant has not, to our knowledge, been published in the literature as pathogenic or benign. Neither BRCA2 c.6415_6416delGAinsAT nor BRCA2 Glu2139Ile (by this or an alternate nucleotide change) was observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, suggesting it is not a common benign variant in these populations. Since Glutamic Acid and Isoleucine differ in polarity, charge, size or other properties, this is considered a non-conservative amino acid substitution. BRCA2 Glu2139Ile occurs at a position that is not conserved and is not located in a functional domain. In silico analyses are inconsistent regarding the effect this variant may have on protein structure and function. Based on currently available information, we consider BRCA2 Glu2139Ile to be a variant of uncertain significance. - |

To our knowledge, no occurrence of c.6415_6416delinsAT in individuals affected with Hereditary Breast And Ovarian Cancer Syndrome and no experimental evidence demonstrating its impact on protein function have been reported. Three clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation.
