BRCA2 (HGNC:1101): (BRCA2 DNA repair associated) Inherited mutations in BRCA1 and this gene, BRCA2, confer increased lifetime risk of developing breast or ovarian cancer. Both BRCA1 and BRCA2 are involved in maintenance of genome stability, specifically the homologous recombination pathway for double-strand DNA repair. The largest exon in both genes is exon 11, which harbors the most important and frequent mutations in breast cancer patients. The BRCA2 gene was found on chromosome 13q12.3 in human. The BRCA2 protein contains several copies of a 70 aa motif called the BRC motif, and these motifs mediate binding to the RAD51 recombinase which functions in DNA repair. BRCA2 is considered a tumor suppressor gene, as tumors with BRCA2 mutations generally exhibit loss of heterozygosity (LOH) of the wild-type allele. [provided by RefSeq, May 2020]

The normal sequence, with the bases that are deleted in braces and inserted in brackets, is TGTT[GA][AT]AGGT. This in frame deletion and insertion, also denoted BRCA2 6643_6644delGAinsAT using alternate nomenclature, occurs on the same allele (in cis) and results in the missense change of a Glutamic Acid to an Isoleucine (GAA>ATA). This variant has not, to our knowledge, been published in the literature as pathogenic or benign. Neither BRCA2 c.6415_6416delGAinsAT nor BRCA2 Glu2139Ile (by this or an alternate nucleotide change) was observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, suggesting it is not a common benign variant in these populations. Since Glutamic Acid and Isoleucine differ in polarity, charge, size or other properties, this is considered a non-conservative amino acid substitution. BRCA2 Glu2139Ile occurs at a position that is not conserved and is not located in a functional domain. In silico analyses are inconsistent regarding the effect this variant may have on protein structure and function. Based on currently available information, we consider BRCA2 Glu2139Ile to be a variant of uncertain significance. -
