Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251400 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.8452G>T has been reported in the literature in individuals from Saudi Arabia, including at least one individual affected with breast cancer (e.g., Abulkhair_2018), as well as a homozygous individual for whom clinical details were not provided, although the individual was reported to have a sibling with leukemia and suspected Fanconi anemia (e.g., AlAbdi_2021, Monies_2023). These reports do not provide unequivocal conclusions about association of the variant with Hereditary Breast And Ovarian Cancer Syndrome.
