This variant was observed as part of a predisposition screen in an ostensibly healthy population. A literature search was performed for the gene, cDNA change, and amino acid change (where applicable). Publications were found based on this search. However, the evidence from the literature, in combination with allele frequency data from public databases where available, was not sufficient to rule this variant in or out of causing disease. Therefore, this variant is classified as a variant of unknown significance. -
Hereditary cancer-predisposing syndrome Pathogenic:1Uncertain:1
This missense variant replaces aspartic acid with glycine at codon 3073 of the BRCA2 protein. Computational prediction suggests that this variant may have deleterious impact on protein structure and function. Functional studies have shown that this variant impacts homology-directed DNA repair activity (PMID: 23108138, 29394989, 33609447) and fails to complement in mouse embryonic stem cells (PMID: 29988080, 33293522) and in human-derived cell lines (PMID: 32444794). This variant has been reported in an individual with a personal or family history of breast and/or ovarian cancer (PMID: 16550498). This variant has been identified in 1/251252 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
The p.D3073G variant (also known as c.9218A>G), located in coding exon 23 of the BRCA2 gene, results from an A to G substitution at nucleotide position 9218. The aspartic acid at codon 3073 is replaced by glycine, an amino acid with similar properties. This alteration is non-functional in multiple assays including multiple homology directed DNA repair assays, a mouse embryonic stem cell survival assay, and multiple drug sensitivity assays (Ambry internal data; Mesman RLS et al. Genet Med, 2019 02;21:293-302; Biswas K et al. NPJ Genom Med, 2020 Dec;5:52; Ikegami M et al. Nat Commun, 2020 05;11:2573). This amino acid position is highly conserved on sequence alignment. This alteration is also predicted to destabilize the local structure and disrupt the protein binding ability of BRCA2 (Yang H et al. Science 2002 Sep;297:1837-48; Marston NJ et al. Mol. Cell. Biol. 1999 Jul;19:4633-42; Ambry internal data). In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the majority of available evidence to date, this variant is likely to be pathogenic. -

This sequence change replaces aspartic acid, which is acidic and polar, with glycine, which is neutral and non-polar, at codon 3073 of the BRCA2 protein (p.Asp3073Gly). This variant is present in population databases (rs80359186, gnomAD 0.0009%). This missense change has been observed in individual(s) with a family history of breast and/or ovarian cancer (PMID: 16550498). ClinVar contains an entry for this variant (Variation ID: 52779). Invitae Evidence Modeling incorporating data from in vitro experimental studies (PMID: 33609447) indicates that this missense variant is expected to disrupt BRCA2 function with a positive predictive value of 95%. Experimental studies have shown that this missense change affects BRCA2 function (PMID: 29394989, 29988080, 33609447). RNA analysis performed to evaluate the impact of this missense change on mRNA splicing indicates it does not significantly alter splicing (internal data). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
