The encoded protein transports ornithine across the inner mitochondrial membrane from the cytosol to the mitochondrial matrix. The protein is an essential component of the urea cycle, and functions in ammonium detoxification and biosynthesis of the amino acid arginine. Mutations in this gene result in hyperornithinemia-hyperammonemia-homocitrullinuria (HHH) syndrome. There is a pseudogene of this locus on the Y chromosome.[provided by RefSeq, May 2009]

Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 4e-06 in 251182 control chromosomes. c.847C>T has been reported in the literature in at least one compound heterozygous individual affected with Hyperornithinemia-Hyperammonemia-Homocitrullinuria Syndrome (e.g., Tessa_2009). These data do not allow any conclusion about variant significance. At least one publication reports experimental evidence evaluating an impact on protein function. The most pronounced variant effect results in 4%-19% of normal activity (Tessa_2009). The following publication has been ascertained in the context of this evaluation (PMID: 19242930).
