This sequence change replaces threonine, which is neutral and polar, with alanine, which is neutral and non-polar, at codon 778 of the RB1 protein (p.Thr778Ala). This variant is present in population databases (no rsID available, gnomAD 0.007%). This variant has not been reported in the literature in individuals affected with RB1-related conditions. ClinVar contains an entry for this variant (Variation ID: 237669). Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Not Available"; PolyPhen-2: "Benign"; Align-GVGD: "Not Available". The alanine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. The p.T778A variant (also known as c.2332A>G), located in coding exon 23 of the RB1 gene, results from an A to G substitution at nucleotide position 2332. The threonine at codon 778 is replaced by alanine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
