Computational prediction tools and conservation analysis do not provide strong support for or against an impact to the protein. Of note, this variant lies in the head region of the protein. Missense variants in this region have been reported and statistically indicated to be more likely to cause disease (Walsh 2016). In addition, two other variants involving this codon, p.Pro307His and p.Pro307Arg, have been identified in individuals with HCM (Mademont-Soler 2017, Coppini 2014, Homburger 2016, Walsh 2017).
