The c.1133T>C (p.I378T) alteration is located in exon 9 (coding exon 7) of the PALS1 gene. This alteration results from a T to C substitution at nucleotide position 1133, causing the isoleucine (I) at amino acid position 378 to be replaced by a threonine (T). This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). This amino acid position is highly conserved in available vertebrate species. The in silico prediction for this alteration is inconclusive. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
