The encoded protein is believed to be an integral membrane protein involved in small molecule transport, specifically tyrosine, which is a precursor to melanin synthesis. It is involved in mammalian pigmentation, where it may control skin color variation and act as a determinant of brown or blue eye color. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with OCA2-related conditions. This variant disrupts a region of the OCA2 protein in which other variant(s) (p.Ala831Pro) have been determined to be pathogenic (PMID: 21458243, 34838614). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing.
