This changes the amino acid from a tryptophan to a stop codon within coding exon 28. This variant was detected in a cohort of early onset complicated hereditary spastic paraplegia (HSP) patients; however, clinical details were limited (Schüle R et al. J Neurol Neurosurg Psychiatry, 2009 Dec;80:1402-4). This alteration has also been reported in the compound heterozygous state in a patient with spastic paraparesis, learning difficulties and a thin corpus callosum (Murugan A et al. Eur J Med.Case Rep, 2017; 1(3):122-125). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. - |
