Variant summary
Our verdict is Pathogenic. Variant got 13 ACMG points: 13P and 0B. PM1PM2PM5PP2PP3_StrongPP5_Moderate
The NM_000138.5(FBN1):āc.4406G>Cā(p.Arg1469Pro) variant causes a missense change involving the alteration of a conserved nucleotide. The variant allele was found at a frequency of 0.000000684 in 1,461,864 control chromosomes in the GnomAD database, with no homozygous occurrence. In-silico tool predicts a pathogenic outcome for this variant. Variant has been reported in ClinVar as Likely pathogenic (ā ). Another variant affecting the same amino acid position, but resulting in a different missense (i.e. R1469C) has been classified as Likely pathogenic.

Marfan syndrome Pathogenic:1
Likely pathogenic, criteria provided, single submitter | clinical testing | Laboratory for Molecular Medicine, Mass General Brigham Personalized Medicine | Oct 13, 2011 | The Arg1469Pro variant has not previously been reported nor previously identifie d by our laboratory in any other families. However, identification of this varia nt in 2 cousins whom have clinical features of Marfan syndrome, and therefore se gregating across 3 informative meioses in this family, increases the likelihood that this variant is pathogenic. In addition, arginine (Arg) at amino acid posit ion 1469 is highly conserved across evolutionarily distant species and this vari ant lies within a functional domain of FBN1, which also increases the likelihood that this variant is pathogenic. In summary, this variant is likely to be patho genic. The clinical significance of this variant should be interpreted in the co ntext of this individual's clinical manifestation. - |
