Other variant(s) that disrupt this residue have been observed in individuals with FBN1-related conditions (PMID: 31730815, 29768367), which suggests that this may be a clinically significant amino acid residue. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant has been observed in individual(s) with clinical features of Marfan syndrome (Invitae). This sequence change replaces glycine with alanine at codon 899 of the FBN1 protein (p.Gly899Ala). The glycine residue is highly conserved and there is a small physicochemical difference between glycine and alanine. This variant is not present in population databases (ExAC no frequency). -
