Variant summary: FBN1 c.156G>A alters a conserved nucleotide resulting in a synonymous change. At least one silico tool predicts benign impact on splicing (TrAP). The variant was absent in 251396 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.156G>A in individuals affected with Marfan Syndrome and no experimental evidence demonstrating its impact on protein function have been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. However, a different nucleotide change at the same position (c.156G>T) resulting in the same amino acid change (p.Ala52=) was classified as benign by 8 laboratories in ClinVar.
