The BTB domain functions as a protein-protein interaction module, which includes an ability to self-associate or to interact with non-BTB domain-containing proteins. The kelch motif typically occurs in groups of five to seven repeats, and has been found in proteins with diverse functions. Known functions of these family members include transcription regulation, ion channel tetramerization and gating, protein ubiquitination or degradation, and cytoskeleton regulation. The cysteine amino acid residue is found in multiple mammalian species, suggesting that this missense change does not adversely affect protein function. These predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This variant has not been reported in the literature in individuals with KBTBD13-related disease. This variant is present in population databases (rs780964425, ExAC 0.08%). This sequence change replaces arginine with cysteine at codon 259 of the KBTBD13 protein (p.Arg259Cys). The arginine residue is weakly conserved and there is a large physicochemical difference between arginine and cysteine. -
