The c.4687_4689delCTCinsTTT variant (also known as p.L1563F), located in coding exon 8 of the ALPK3 gene, results from an in-frame deletion of CTC and insertion of TTT at nucleotide positions 4687 to 4689. This results in the substitution of the leucine residue for a phenylalanine residue at codon 1563, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be inconclusive by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
