Variant summary: POLG c.1613A>C (p.Glu538Ala) results in a non-conservative amino acid change in the encoded protein sequence. Three of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 0.00012 in 251370 control chromosomes, predominantly at a frequency of 0.00087 within the Latino subpopulation in the gnomAD database. To our knowledge, no occurrence of c.1613A>C in individuals affected with POLG-Related Spectrum Disorders and no experimental evidence demonstrating its impact on protein function have been reported. Four submitters have cited clinical-significance assessments for this variant to ClinVar after 2014. All submitters classified the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance. -

The c.1613A>C (p.E538A) alteration is located in exon 9 (coding exon 8) of the POLG gene. This alteration results from a A to C substitution at nucleotide position 1613, causing the glutamic acid (E) at amino acid position 538 to be replaced by an alanine (A). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -

This sequence change replaces glutamic acid, which is acidic and polar, with alanine, which is neutral and non-polar, at codon 538 of the POLG protein (p.Glu538Ala). This variant is present in population databases (rs767216577, gnomAD 0.09%). This variant has not been reported in the literature in individuals affected with POLG-related conditions. ClinVar contains an entry for this variant (Variation ID: 432020). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant.
