Genes affected
BLM (HGNC:1058): (BLM RecQ like helicase) The Bloom syndrome is an autosomal recessive disorder characterized by growth deficiency, microcephaly and immunodeficiency among others. It is caused by homozygous or compound heterozygous mutation in the gene encoding DNA helicase RecQ protein on chromosome 15q26. This Bloom-associated helicase unwinds a variety of DNA substrates including Holliday junction, and is involved in several pathways contributing to the maintenance of genome stability. Identification of pathogenic Bloom variants is required for heterozygote testing in at-risk families. [provided by RefSeq, May 2020]

Uncertain significance, criteria provided, single submitter | clinical testing | Ambry Genetics | Jul 15, 2022 | The p.R1139P variant (also known as c.3416G>C), located in coding exon 17 of the BLM gene, results from a G to C substitution at nucleotide position 3416. The arginine at codon 1139 is replaced by proline, an amino acid with dissimilar properties. This alteration has been reported in individuals diagnosed with breast cancer (Sokolenko AP et al. Int. J. Cancer 2012 Jun;130:2867-73; Sokolenko AP et al. Cancer Lett. 2015 Apr;359:259-61). This alteration was also reported in 1/107 Macedonian individuals with a clinical history of hereditary polyposis or hereditary non-polyposis colorectal cancer who underwent multi-gene panel testing (Staninova-Stojovska M et al. Balkan J Med Genet, 2019 Dec;22:5-16). Additionally, this alteration has been reported in 1/1197 individuals from Greece, Romania, and Turkey undergoing evaluation for inherited cancer predisposition (Tsaousis GN et al. BMC Cancer, 2019 Jun;19:535). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
