TSC2 (HGNC:12363): (TSC complex subunit 2) This gene is a tumor suppressor gene that encodes the growth inhibitory protein tuberin. Tuberin interacts with hamartin to form the TSC protein complex which functions in the control of cell growth. This TSC protein complex negatively regulates mammalian target of rapamycin complex 1 (mTORC1) signaling which is a major regulator of anabolic cell growth. Mutations in this gene have been associated with tuberous sclerosis and lymphangioleiomyomatosis. [provided by RefSeq, May 2022]

Variant summary: TSC2 c.4870_4871delCT (p.Leu1624AspfsX28) results in a premature termination codon, predicted to cause absence of the protein due to nonsense mediated decay, which is a commonly known mechanism for disease. The variant was absent in 249492 control chromosomes. c.4870_4871delCT has been reported in the literature in at-least one individual affected with Tuberous Sclerosis Complex (Niida_1999). To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication has been ascertained in the context of this evaluation (PMID: 10533067). ClinVar contains an entry for this variant (Variation ID: 49329). Based on the evidence outlined above, the variant was classified as pathogenic.
