TBC domain proteins may serve as GTPase-activating proteins for a particular group of GTPases, the Rab (Ras-related proteins in brain) small GTPases which are involved in the regulation of membrane trafficking. p.Gly110Gly in exon 2 of TBC1D24: This variant is not expected to have clinical significance because it does not alter an amino acid residue and is not located within the splice consensus sequence. It has been identified in 3/8496 of East A sian chromosomes by the Exome Aggregation Consortium (ExAC, http://exac.broadins titute.org). Splicing computational tools predict the possible creation of a nov el splice site, but the accuracy of these tools is unknown. Although a role in d isease cannot be fully excluded, this variant is more likely benign. -
