Variant summary: CYBA c.74G>A (p.Gly25Asp) results in a non-conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The frequency data for this variant in gnomAD is considered unreliable, as metrics indicate poor data quality at this position. c.74G>A has been reported in the literature as a homozygous genotype in at-least one individual affected with Chronic Granulomatous Disease (example, Koker_2013 cited in Roos_2021, Akar_2021). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
