Variant summary: FANCA c.1480C>G (p.Leu494Val) results in a conservative amino acid change located in the Fanconi anaemia group A protein, N-terminal domain (IPR031729) of the encoded protein sequence. Three of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251352 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.1480C>G has been reported in the literature in an individual affected with Fanconi Anemia (Wang_2023). These data do not allow any conclusion about variant significance. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication has been ascertained in the context of this evaluation (PMID: 36513378). No submitters have cited clinical-significance assessments for this variant to ClinVar. The FANCA c.1480C>G (p.Leu494Val) variant has been reported in the published literature in an adult-age individual with Fanconi anemia who carried a pathogenic variant in the FANCA gene presumably on the opposite chromosome (PMID: 36513378 (2023)). This variant has not been reported in large, multi-ethnic general populations (Genome Aggregation Database, http://gnomad.broadinstitute.org). Analysis of this variant using bioinformatics tools for the prediction of the effect of amino acid changes on protein structure and function yielded conflicting predictions that this variant is benign or damaging. Based on the available information, we are unable to determine the clinical significance of this variant. -
