Other variant(s) that disrupt this residue have been observed in individuals with PMP22-related conditions (PMID: 21337347), which suggests that this may be a clinically significant amino acid residue. An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. ClinVar contains an entry for this variant (Variation ID: 570254). This missense change has been observed in individual(s) with clinical features of PMP22-related conditions (Invitae). This variant is not present in population databases (gnomAD no frequency). This sequence change replaces arginine, which is basic and polar, with histidine, which is basic and polar, at codon 159 of the PMP22 protein (p.Arg159His). -
