The asparagine at codon 746 is replaced by serine, an amino acid with highly similar properties. This alteration was identified as a VUS in a cohort of 524 unselected Chinese breast cancer patients who underwent multi-gene panel testing (Chen B et al. Aging (Albany NY), 2020 02;12:3140-3155). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
