ClinVar contains an entry for this variant (Variation ID: 457573). The p.N746S variant (also known as c.2237A>G), located in coding exon 18 of the NF1 gene, results from an A to G substitution at nucleotide position 2237. The asparagine at codon 746 is replaced by serine, an amino acid with highly similar properties. This alteration was identified as a VUS in a cohort of 524 unselected Chinese breast cancer patients who underwent multi-gene panel testing (Chen B et al. Aging (Albany NY), 2020 02;12:3140-3155). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
