They are responsible for the generation and propagation of action potentials in neurons and muscle. This gene encodes one member of the sodium channel alpha subunit gene family. This variant has been observed in combination with a different SCN4A variant in a family with congenital myopathy (PMID: 30369941). ClinVar contains an entry for this variant (Variation ID: 586523). This variant is present in population databases (rs774183791, ExAC 0.001%). This sequence change replaces proline with leucine at codon 1650 of the SCN4A protein (p.Pro1650Leu). The proline residue is highly conserved and there is a moderate physicochemical difference between proline and leucine. -
